I’ve always tried to refer to people, present or not, as whatever gender they seem to want to present.
I was taught years ago to use “she” for even the most obvious drag queens and transvestites, as a courtesy… and these days many articles about people note their preferred “pronouns”, whether they’re trans or even cis-gendered.
I’m kind of surprised to hear Jane call Ted “she”, especially when admitting to seeing no evidence.

I’m still new to this comic, I’m sometimes confused by who’s male or female. Also, dig the mitt on Jane, I wouldn’t want to get smacked by her.

Ted is ambiguous except in the character department. Jerk is the euphemism here.
